引言
囊胚互补通过使用多能干细胞(PSCs)来补充缺乏器官发育能力的胚胎,从而帮助解决器官短缺的问题【1】。然而,在进化距离较远的物种中应用这一技术时,往往会遇到细胞竞争和发育时间差异等异种屏障的挑战【2-3】。
2024年8月23日,来自美国得克萨斯州大学西南医学中心分子生物系的吴军团队在Cell Stem Cell期刊发表题为Incompatibility in cell adhesion constitutes a barrier to interspecies chimerism(细胞粘附的不兼容性构成了异种嵌合体的形成障碍)的文章。研究发现,远缘物种多能干细胞之间的粘附不兼容性是异种嵌合体形成的主要障碍。为了克服这一问题,他们开发了一种合成纳米抗体介导的粘附策略,成功增强了人类细胞在异种嵌合体中的贡献。
